ABSTRACT

Objective To establish a HPLC fingerprints of Biyuanjing capsules. Methods The column was Agilent SB-C18(4.6mm×250 mm, 5 µm). The mobile phase was acetonitrile-water with gradient elution at a flow rate of 1.0 ml/min. The detection wavelength was 210 nm. The detection time was 80 min. Results The HPLC fingerprints of Biyuanjing capsules were established. Twenty common peaks were confirmed, of which, 15 peaks were belonging to each crude drug and 5 peaks were identified as chemical components. The overall similarity of the fingerprints of 10 batches of samples was above 90% comparing with the control. Conclusion This method can be used for the quality control of Biyuanjing capsules.

ABSTRACT

Objective: To study the secondary metabolites of endophytic fungus Aspergillus ochraceus SX-C7 from Setaginella stauntoniana. Methods: The compounds were isolated and purified by silica gel, Sephadex LH-20, C-18 reversed phase column chromatography, and their structures were identified by MS and NMR analyses. Antibacterial activity and inhibitory activity against acetylcholinesterase were studied by doubling dilution and Ellman methods. Results: Ten known compounds were isolated from the secondary metabolites of A. ochrateus SX-C7, and identified as: semivioxanthin (1), 6-hydroxy-p-menth-4 (5)-en-3-one (2), flavacol (3), magnolin (4), preussin B (5), circumdatins D (6), isofucosterol (7), sclerotiamide (8), 3β-hydroxyergosta-8,24(28)-dien-7-one (9), and 3-O-β-D-glucopyranosyl stigmasta-5(6),24(28)-diene (10). Conclusion: Compounds 1, 2, 4, 7, and 10 were isolated from A. ochraceus SX-C7 for the first time. The results of the bioactivity assay showed that compound 10 possessed obvious inhibitory activity against Bacillus subtilis with a MIC value of 2 μg/mL, which was at the same grade with positive control. Compound 1 exhibited potent inhibitory activity against acetylcholinesterase with an inhibitory rate of 62.3% (the final concentration was 50 μmol/L.

ABSTRACT

Objective: To study the chemical constituents from the branches and leaves of Viburnum sargentii collected at Mountain Tai. Methods: The chemical constituents were isolated and purified by chromatographic methods, including silica gel, ODS, Sephadex LH-20 columns, and RP HPLC. The structures of the isolated compounds were identified by ESI-MS, 1D-NMR, and 2D-NMR data analyses, and compared with the literature data. Results: Thirteen compounds were obtained from the 95% ethanol extract of branches and the leaves of V. sargentii, and determined as α-amyrin (1), uvaol (2), 3α-ursolic acid (3), 11,12-dehydroursolic acid lactone (4), 3-O-acetyloleanolic aldehyde (5), oleanolic acid (6), betulinic acid (7), magnolin (8), (+)-eudesmin (9), (-)-epieudesmin (10), vibsanol (11), 3,4'-dimethoxylvibsanol (12), and α-asarone (13), respectively. Conclusion: Compound 12 (3,4'- dimethoxylvibsanol) is a new natural product, and compounds 1-11 and 13 are isolated from V. sargentii for the first time.

ABSTRACT

OBJECTIVE:To study the pharmacokinetics of magnolin in rats. METHODS:High performance liquid chromatog-raphy(HPLC)was adopted. The determination was performed on Kromasil C18 with mobile phase consisted of acetonitrile-tetrahy-drofuran-water (39∶1∶60),at the flow rate of 1.0 ml/min,with the measurement wavelength of 278 nm,column temperature of 35 ℃ and the sample size of 20 μl. 8 Wistar rats were docked to collect blood from the caudal vein before administration [10 mg(medicinal materials)/kg] and 0.25,0.5,0.75,1,1.5,2,4,8,12 and 20 h after administration,to determine the concentra-tion of the drug in blood. DAS2.1.1 software was used to calculate pharmacokinetic parameters. RESULTS:In the determination of magnolin,the mass concentration linear range was found to be 0.05-10.00 μg/ml(r=0.999 5);the RSD of precision test and sta-bility test were less than 13%(n=6);relative recovery rate was 97.32%-102.15%(n=6);extraction recovery rate was 84.63%-90.02%;t1/2α of magnolin in rats was(0.48±0.22)h,t1/2β was(7.96±2.57)h,CL/F was(0.09±0.032)L/(h·kg),and AUC0-20 h was(944.43±212.83)mg·h/L. CONCLUSIONS:This method conforms to the requirements for the determination of bio-logical samples with respect to precision,stability and accuracy. Magnolin demonstrates a good linear relationship between AUC0-20 h and the dose in rats,with the process compatible with two-compartment model.

ABSTRACT

Objective:To develop a method to simultaneously determine the content of imperatorin, isoimperatorin and magnolin in Quchuan capsules by HPLC. Methods:A Shiseido CAPCELL PAK C18 column(150 mm × 4. 6 mm,5 μm) was used. The mobile phase consisted of acetonitrile(A)-water(B) with gradient elution at a flow rate of 1. 0 ml·min-1 and the column temperature was at 30℃. The detection wavelength was set at 278 nm from 0 to 25 min and 248 nm from 25 to 70 min. The injection volume was 10 μl. Results:The linear range of imperatorin,isoimperatorin and magnolin was 6. 126-55. 134 μg·ml-1(r=0. 999 7), 9. 862-88. 758 μg ·ml-1(r =0.999 8) and 4.830-43.470 μg·ml-1(r =0.999 8), respectively. The average recovery was 101.5% (RSD =1. 4%), 99. 6%(RSD=1. 2%) and 100. 2% (RSD=1. 7%), respectively. Conclusion: The method is convenient, sensitive and accurate, which can be used for the quality control of Quchuan capsules.

ABSTRACT

Objective To develop a method for determination of magnolin in Bimingan Pellets. Methods HPLC was adopted for determination by using Agilent Eclipse XDB-C18 column (4.6 mm×150 mm, 5 μm), acetonitrile-water(40∶60) as the mobile phase. The detection wavelength was 278 nm. Results The method was linear in the range of 0.117~2.340 μg for the injection volume of magnolin(r=0.999 94), and the average recovery was 99.4%, RSD=1.9%. Conclusion The method was simple, accurate, reliable and reproducible for the quality control of Bimingan Pellets.

ABSTRACT

BACKGROUND/AIMS: There is a need for new anti-asthmatic medications with fewer side effects. NDC-052, an extract of the medicinal herb Magnoliae flos, which has a long history of clinical use, was recently found to have anti-inflammatory effects. Herein, we evaluated the effects of NDC-052 as an add-on therapy in patients with mild to moderate asthma using inhaled corticosteroids (ICS). METHODS: In a non-comparative, multi-center trial, 148 patients taking ICS received NDC-052 for eight weeks. We evaluated their forced expiratory volume in one second (FEV1), morning and evening peak expiratory flow rate (AM and PM PEFR), AM/PM asthma symptom scores, visual analogue symptom (VAS) scores, night-time wakening, frequency of short-acting beta2-agonist usage, and adverse events. RESULTS: After eight weeks, both AM and PM PEFRs were significantly improved. Asthma symptom scores, VAS scores, the frequency of nights without awakening, and the frequency of beta2-agonist use were also reduced. Most of the adverse drug reactions were mild and resolved spontaneously. CONCLUSIONS: The addition of NDC-052 to ICS had a beneficial effect on asthma control in patients with mild to moderate asthma, with good tolerability and fewer side effects. Further studies are necessary to evaluate the effects of NDC-052 in patients with severe and/or refractory asthma.

ABSTRACT

Objective To establish a HPLC method for the determination of magnolin in Xuetie Dingchuan Plaster.Methods The HPLC chromatographic condition was as follows: the analytical column was Lichrospher C8(4.6mm?250mm,5?m),the mobile phase consisted of A(acetonitrile)∶B(1.55%tetrahydrof uran-water) 34∶66,the flow rate was 1.0 mL?min-1,the detected wav elength was 278 nm,and the column temperature was 30 ℃.Results The calibr ation curve was linear(r=0.9999) in the range of 0.42~ 5.25 ?g for magnolin,the average recovery of the method was 100.50 %,with RSD 2.22 %(n=9).Conclusion The method is accurate,simple and stable,and can be used for the quality control of Xuetie Dingchuan Plaster.

ABSTRACT

Object To investigate the process of flocculation impurity removal for the Anti-flu Nose Drops (AND). Methods Five kind of effective factors on flocculation impurity removal with the index of clarifying results were studied. Comparative tests for Shandong Yaosheng 1 + 1 (Ⅲ) clarificant, 50% alcohol precipitation, and 75% alcohol precipitation were performed with the quantitative index of gardenoside contents, the qualitative index of magnolin and gardenoside TLC chromatograms. The optimum process condition was selected out. Results Under the optimum process condition of flocculation impurity removal, gardenoside contents in the flocculation group is higher than those in 50% , 75% alcohol groups. TLC chromatograms showed that the spots of flocculation group are the same as those in the original liquid group. Conclusion Shandong Yaosheng 1 + 1 (Ⅲ) clarificant has following advantages; high reserved rate of active components, simple operation, cheap cost, stable process, which is a ideal method for the impurity removal.
